Abstract
甲殼類與軟體動物為主要的食品過敏原來源,而具永續性的食用昆蟲亦具有潛在的致敏風險。甲殼類、軟體動物及昆蟲同屬於無脊椎動物,其共同的主要致敏性蛋白質為原肌球蛋白 (tropomyosin, TM)。由於原肌球蛋白在結構上具有高度穩定性,其於食品加工過程中的致敏性變化機制仍未被充分釐清,特別是酸、鹼與酵素處理對其免疫反應性影響的相關研究仍相當有限。因此本研究以甲殼類、軟體動物及食用昆蟲之原肌球蛋白作為研究標的,第一步比較酸處理、鹼處理與酵素處理對其免疫反應性的影響。研究結果顯示,酸處理能明顯改變原肌球蛋白的免疫反應性,因此本研究進一步聚焦於酸處理對無脊椎動物致敏性蛋白質原肌球蛋白之影響。實驗中以白醋作為酸處理來源,選用白蝦(甲殼類)、花枝(軟體動物)、白蟋蟀與麵包蟲(食用昆蟲)作為代表性樣本。在處理時間試驗中,經白醋處理15分鐘即可使各類樣本中原肌球蛋白的免疫反應性明顯下降;而在濃度試驗中,以15% 白醋處理1小時同樣能有效降低其免疫反應性。進一步以9種甲殼類、軟體動物及食用昆蟲物種進行擴大驗證,結果顯示酸處理皆能降低原肌球蛋白的免疫反應性,顯示其具有跨物種的一致性。基於研究結果,酸處理可改變無脊椎動物原肌球蛋白的免疫反應性表現,推測其可能與蛋白質構型及抗原表位改變有關,並為後續探討原肌球蛋白於酸性環境下之檢測特性與致敏性調控提供重要的基礎資訊。
