Abstract
能量代謝最主要胞器為粒線體,近年來,許多藥物或保健食品相關研究,皆注重粒線體調控能量代謝的作用。論文主軸為高脂飲食或致癌物誘發不同層次粒線體受損動物模式,可能機制則利用細胞實驗確立。研究一,非酒精性脂肪肝易引起脂肪毒性、粒線體功能受損和氧化壓力。α- mangostin為山竹果皮中主要多酚成分,已被證實可改善脂肪肝及肥胖
,但其粒線體相關機制尚未明確。因此,本研究目的富α-mangostin的山竹萃取物對高脂飲食誘發脂肪肝的大鼠之影響。結果顯示,高脂飲食引起大鼠肝性脂肪變性,導致肝臟中粒線體功能受損和粒線體依賴行細胞凋亡的產生,給予α-mangostin可藉由抗氧化物作用恢復粒線體功能正常。研究二,前列腺癌細胞主要為有氧糖解路徑產能,粒線體功能幾乎無功能。近年來臨床研究發現,metformin可降低前列腺癌的發生率,然而metformin為能量代謝促進劑,其是否與粒線體有關的研究尚未清楚。因此,本研究目的,給予metformin其對可前列腺癌能量代謝之探討。結果顯示,給予metformin可藉由活化AMPK (adenosine 5‘-monophosphate (AMP)-activated protein kinase) 啟動粒線體氧化磷酸化 (oxidative phosphorylation, OXPHOS) 功能。本研究結論,維持粒線體生物能量功能及代謝可能改善粒線體功能受損疾病,如前列腺癌及非酒精性脂肪肝。
