Abstract
海洋覆蓋了地球70%以上的表面積,富有許多值得開發的天然資源。Sinularia flexibilis 是一種軟珊瑚,由於缺乏外骨骼,軟珊瑚通常會透過化學防禦的方式產生次級代謝物來保護自身不受掠食者侵害,而這些具有生物活性的次級代謝物則為新藥開發之重要起源。本篇論文旨於探討在不同生長環境下柔軟指形珊瑚S. flexibilis所產生的次級代謝物差異。首先,應用超高效液相色譜結合四極桿飛行時間質譜獲得所有樣品的MS和MS/MS數據。接續以主成分分析 (Principal component analysis, PCA) 與正交偏最小二乘判別分析 (Orthogonal projections to latent structures discriminant analysis, OPLS-DA)針對所有代謝物進行差異分析,結果顯示野生與養殖珊瑚間存在顯著差異。進一步使用 MS/MS 數據構築分子網絡 (Molecular networking, MN) 圖譜。視覺化的分子網絡圖譜顯示,養殖型 S. flexibilis 軟珊瑚的化學結構多樣性較高,尤其含有較多的 diterpenoids和diterpene dimers骨架化合物。總結以上,由於誘發化學防禦的刺激來源差異,不同來源或不同生長環境的軟珊瑚表現的次級代謝物亦存在一定的差異,因此進一步尋找此等關鍵次級代謝物含量的變因,是將來養殖型珊瑚作為藥用成分量產的重要課題。
