Abstract
前列腺位於盆底和尿道周圍區域,也是男性最大的性附屬器官。它主要作 用於精液的產生、射精時尿道的閉合和內分泌代謝。當前列腺的異常細胞 若以不受控制的方式生長時,就會發生前列腺癌 (PC),從而導致惡性腫瘤的形成。 前列腺癌的病理學是高度複雜的,它是由各種途徑和分子事件導致的,其涉及炎症、免疫反應和癌症基因在起始和惡化進展過程。至今,科學家仍在探索這些機制以用於篩選及確定治療前列腺癌治療效果的適當生物標誌物。因此,我們旨在建立一個臨床試驗案,旨在使用來自前列腺患者血液樣本的潛在生物標誌物候選物來檢測快速進展的前列腺癌。 迄今為止,前列腺特異性抗原(PSA)是最可靠的前列腺生物標誌物;然而,在正常衰老、良性前列腺增生(前列腺肥大)和前列腺炎(前列腺炎症)等各種其他情況下,PSA 檢測值會高於正常標準(4 ng/mL),因此PSA 生物標誌存在偽陽性風險。在前列腺患者中也觀察到低於 4 ng/mL 的前列腺特異性抗原值,這影響了生物標誌物的可靠性。此外,前列腺特異性 抗原在無症狀男性篩檢前列腺癌中的作用仍存在爭議。因此,根據文獻回顧,我們篩選了13種生物標誌物,它們可能是前列腺進展特異性的潛在候選物。這些生物標誌物包括 PSA、TMPRSS2-ERG、PCA3、AR-V7、SLIT2、PNPLA7、L1CAM、LIF、CHRM4、sADAM9、LGALS7B、ZNF253 和 ADRA1D。最後,我們預計建立我們的預測前列腺癌快速進展的特異性生物標誌物,有助於早期檢測前列腺以實施適當的治療.
