Abstract
樹突狀細胞(dentritic cell)是一種抗原呈現細胞,其在自然免疫與適應性免疫搭起了溝通的橋樑,且為啟動整個免疫反應的關鍵。而在樹突狀細胞上的兩種受器Toll-like receptors及 C-type lectin receptors在抗原辨識與免疫啟動扮演著非常重要的角色。其中C-type lectin receptors 在與一些特殊的醣分子結構結合時可以誘發不同的免疫反應。而目前過敏已成為一個嚴重的小兒健康問題,並且又以氣喘及食物過敏最為常見。其被認為與第二型輔助型T細胞與免疫球蛋白IgE的上升有關。代表了過敏的病人無法成功對過敏原產生耐受性。我們的實驗發現塵蟎及百慕達草這兩種過敏原的醣類分子是與樹突狀細胞上的C-type lectin receptors 結合的關鍵,而這種過敏原及受器的結合可以引起數突狀細胞內的Raf-1的訊息傳導並磷酸化NF-κB的次單元p65, p65可做為一轉錄因子而引起基因的轉錄,尤其是腫瘤壞死因子,TNF-α.但是在不同個體的人所取出而培養的樹突狀細胞在面對塵蟎的刺激時,有人可以產生上面描述的反應,而有的人不行,這可能與免疫系統產生免疫反應及耐受性有關。而在我們的mRNA表現上的實驗可發現,在所有接受塵蟎刺激的個體皆可產生osteopontin,而且在免疫上有調節發炎及抑制發炎的雙面功能,這也許就可以解釋為什麼塵蟎刺激的不同個體會有不同的反應,有的人走向發炎過敏,有人的卻走向抑制過敏發炎,也代表了免疫反應及耐受性的產生。
