Abstract
在胚胎發育初期,Satb2 (Special AT-rich sequence-binding protein 2) 蛋白已知表 現於胚胎幹細胞並影響後續細胞命運。而在生殖細胞系的發育過程中,原始生殖細胞 (primordial germ cells, PGCs) 會藉由多種訊號的影響遷徙到生殖脊並發育為生殖幹細胞 (germline stem cells, GSCs)。在此過程中,Satb2 在 PGCs 與 GSCs 的表現具有顯著差異 性,但我們目前仍不清楚 Satb2 在生殖細胞所扮演的精確功能。為了確保小鼠生殖細胞 發育正確,生殖細胞在胚胎日齡 E6.0 至 E12.5 中經歷三個關鍵事件,包括抑制體細胞 程序、重新獲得潛在的多能性和全基因組表觀遺傳重編程, 然後經過性別分化後成為生 殖母細胞 (gonocytes),再形成雄性的精原幹細胞或雌性的卵母細胞中。過去的研究發現 Satb2 在胚胎發育中作為轉錄因子和染色質重塑蛋白並參與了基因表達、細胞多能性和 表觀遺傳的調控。在本論文中,我們首重探討 Satb2 在小鼠生殖細胞發育中的功能和 表達趨勢, 分析 Satb2 在胚胎日齡 E10.5 至 E15.5 小鼠生殖細胞中的時間與空間分佈 和可能的分子機制。免疫組織螢光染色結果顯示 Satb2在 E10.5(data?)至E12.5的PGCs 中表達,然後在 E13.5~E15.5 的 gonocyte 裡降低表現,說明 Satb2 在原始生殖細胞與生 殖母細胞的發育中有顯著的差異性調控。 另外,在體外無血清培養系統中培養生殖幹細胞時可發現,缺少Satb2 的生殖細胞顯著降低了鹼性磷酸酶活性、多能性相關基因表現量(Oct4、Sox2、Nanog、Klf-4)和細胞增殖相關的 Igf-1r 蛋白質的表達。其中若 以 Igf-1 刺激體外培養的生殖幹細胞,可增加 Satb2、Igf-1r 和 Oct4 蛋白的表達。總 括而言,目前的研究結果表明 Satb2 在不同時期的胚胎存在差異性調控機制,且其中一 條調控路徑可能透過 Igf-1/Igf-1r 信號通路來調節 GSCs 的生物功能性。
