Abstract
MCP-induced protein 1 (MCPIP1又被稱為ZC3H12A) 為鋅指蛋白家族成員之一,最早是在MCP-1誘導的人類周邊單核球中被發現到。先前的文獻中發現,MCPIP1參與在脂肪生成、血管新生及發炎作用。目前的文獻推測MCPIP1在所具有的功能可能與具有RNase的功能。
本研究使用可藉由doxycycline (Tet/On system)誘導表現MCPIP1的細胞株 (TRex-hek293),分別可表現野生型MCPIP1及突變型MCPIP1 (PIN domain突變),欲探討MCPIP1在細胞生長中所扮演角色及其分子機制為何。
經MTT分析我們發現野生型MCPIP1的表現可以抑制細胞生長,並且在顯微鏡觀察下,發現MCPIP1可以使細胞外觀變圓而且貼附能力下降。然後經LDH釋放分析和annexin V-FITC/PI雙染流式細胞儀分析下發現,長時間表現MCPIP1的細胞雖然會造成生長抑制但是細胞卻沒有伴隨嚴重的細胞毒性現象,顯示MCPIP1具有cytostatic的作用。
同時,結果也顯示MCPIP1不僅僅造成細胞週期蛋白在蛋白層次改變外,在RNA層次上也會受到影響。顯示MCPIP1所造成抑制生長的表現是藉由影響細胞週期所造成。
以上結果顯示出MCPIP1的表現可能是藉由MCPIP1 PIN domain的來影響細胞週期的調控蛋白來達到細胞抑制生長的結果。
