Abstract
異位性皮膚炎是一種發炎性皮膚病,表皮的發炎致使皮膚嚴重的搔癢降低患者的生活品質。此外,患者增加的Tumor necrosis factor-alpha (TNF-α)和Interferon-gamma (IFN-γ)等細胞激素會引起免疫浸潤,引發角質細胞凋亡,致使病情惡化。然而,現今許多療法卻會產生嚴重的副作用。為了增加臨床效益,我們探討檜木醇 (Hinokitiol)其對異位性皮膚炎之發炎及細胞凋亡的影響。使用 2,4-二硝基氯苯 (2,4-Dinitrochlorobenzene, DNCB)塗抹動物皮膚,建立異位性皮膚炎動物模式 ,檜木醇可以恢復 DNCB 誘導的皮膚和耳朵表皮和真皮層厚度,調節Toll-like receptor 4/ Myeloid differentiation primary response protein 88 (TLR4/ MyD88) 通路,降低發炎反應。在細胞模式中,TNF-α和IFN-γ會導致HaCaT細胞凋亡,而檜木醇可以保護細胞免於Reactive oxygen species (ROS)損傷,降低細胞凋亡相關蛋白的表現,提高其緊密連接蛋白的表現。本研究為首篇探討檜木醇對異位性皮膚炎之發炎和細胞凋亡之研究,旨在改善異位性皮膚炎的狀況。結果顯示,檜木醇可以作為異位性皮膚炎的輔助療法。
