Abstract
飲食中多元不飽和脂肪酸(PUFA)的組成被認為可能與發炎相關慢性疾病的發生相關。研究指出給予較多n-3 PUFA可改善實驗動物脂質代謝情形;而腸道菌相的改變與能量代謝異常亦為造成糖尿病(DM)發生的主要風險因子,因此推測腸道菌相的改變可能與DM的發生有著互為因果關聯性。本研究先餵食SD大鼠高脂飼料,再給予STZ (30 mg/kg i.p.)誘發產生糖尿病症狀;誘發成功後再調整飼料中n-6/n-3 PUFA比例,分別為:Regular組(R)、Low組(L)及High組(H),其n-6/n-3 PUFA比值依序為6.5、3.0、9.4,以探討不同PUFA比例是否可以改善糖尿病之代謝相關因子。並以次世代基因定序(NGS)探討糖尿病的發生及飼料介入對於腸道菌相是否有影響。實驗結果顯示在誘發糖尿病症狀下,高n-6/n-3 PUFA比例飼料,會提高糖尿病大鼠體內的氧化壓力指標丙二醛(MDA)之濃度,甚至使腎臟中IL-6的濃度顯著增加。相對而言,糖尿病大鼠攝食低n-6/n-3 PUFA比例飼料6週後,可顯著穩定血糖,對大鼠之血糖具有正面改善的效應,但對體內之循環發炎指標TNF-α並沒有改善;此外在糖尿病疾病狀態下,厚壁菌門(Firmicutes)數量有下降的趨勢。因此推論低比例n-6/n-3有改善血糖恆定之效果,組織發炎反應及腸道菌相的歧異度相關性較低。
