Abstract
本研究旨在探討大豆抗性蛋白 (soy resistant protein, SRP) 對於非酒精性脂肪肝臟疾病 (nonalcoholic fatty liver disease, NAFLD) 之影響,並探討其相關機制。實驗共進行 18 週,實驗初期給予 8 週齡雄性 SD 大鼠 12 週 AIN-93M 液態標準飲食或 Lieber-Decarli 液態高脂飲食,以建立大鼠NAFLD疾病模式,再將實驗動物分為四組,分別為控制組 (C)、NAFLD 組 (N)、NAFLD + 12.5% SRP 組 (NSL) 及 NAFLD + 25% SRP 組 (NSH),並開始為期6週的介入期,介入組飲食以 SRP 取代部分酪蛋白。實驗期間記錄體重及攝食量變化,並測量血液生化數值;實驗末採集血液、肝臟、脂肪組織及盲腸內容物進行分析。實驗結果顯示:高脂飲食成功誘導大鼠產生肝功能損傷、脂肪堆積及發炎情形;而 SRP 的介入顯著改善肝功能指數上升,並有減緩胰島素阻抗的趨勢;且透過病理切片結果也觀察到有助於改善肝臟脂質堆積及發炎情形,亦可顯著降低肝臟三酸甘油酯及膽固醇濃度,同時也具有改善肝臟 MDA 及細胞發炎激素 TNF-α、IL-6 及 IL-1β 濃度的趨勢;此外,在腸道菌項分析結果方面顯示大豆抗性蛋白能顯著改變腸道菌相組成,並且 Spearman 相關性分析顯示菌相改變與肝臟相關生物指標具有關聯性。因此,SRP 藉由改善肝臟脂質堆積、氧化壓力及發炎反應,並可能透過影響腸道菌叢,達到改善 NAFLD 疾病進程的發展。
