Abstract
非酒精性脂肪肝疾病 (non-alcoholic fatty liver disease, NAFLD) 為全球主要的肝臟疾病之一,而 cafeteria 飲食為一項以人類零食誘導動物產生 NAFLD 的飲食模式,目的為更貼近西化飲食對健康的影響。近年來,為提升農業副產物之利用率,發酵米糠之相關研究指出其中含量豐富的 γ-胺基丁酸 (γ-aminobutyric acid, GABA) 被視為具有改善代謝性疾病潛力之關鍵成分。因此,本實驗旨在以脂肪-腸-肝軸之觀點探討富含 GABA 米糠發酵物對臺灣形式 cafeteria 飲食誘導大鼠 NAFLD 之預防效果。本實驗為期 16 週,將 32 隻 5 週齡雄性 Wistar 大鼠分為四組:正常飲食組 (C)、正常飲食補充 300 mg/kg BW 富含 GABA 米糠發酵物組 (CG)、cafeteria 飲食組 (CAF) 及cafeteria 飲食補充 300 mg/kg BW 富含 GABA 米糠發酵物組 (CAFG)。16 週 cafeteria飲食顯著誘導大鼠出現肥胖、血漿與肝臟三酸甘油酯濃度上升、胰島素阻抗、肝臟脂肪堆積、發炎與氧化壓力增加、NAFLD score 上升、脂肪細胞體積增加、瘦素與脂肪激素分泌增加;同時使菌相多樣性下降、Lactobacillus 和 Allobaculum 豐度增加、結腸屏障受損,以及糞便乙酸與丁酸濃度下降。每日餵食 300 mg/kg BW 之富含GABA 米糠發酵物可降低體重、血漿三酸甘油酯濃度及 NAFLD score,並提高菌相多樣性與 Muribaculaceae 和 Roseburia 豐度,顯示其對 NAFLD 之潛在預防效果。
