Abstract
大腸癌為全球癌症相關死亡的主要原因之一,其癌變的過程被認為是正常細胞中基因變異及表觀遺傳變化積累的結果。在表觀遺傳修飾中,組蛋白甲基轉移酶 (HMT) 在腫瘤發生進程的表觀遺傳變異中之關鍵作用已被廣泛報導。癌症相關基因上的異常組蛋白甲基化會導致癌症進展,其機制包括癌症的發生、增殖、轉移和耐藥性的出現。靶向組蛋白甲基轉移酶作為抗癌治療策略引起了相當大的關注,然而,僅有少數組蛋白甲基轉移酶抑制劑能在臨床領域取得成功。因此,我們的研究目標是開發組蛋白甲基轉移酶抑制劑的小分子藥物作為潛在癌症治療方法,並探討藥物對癌細胞株的生物學影響。 在這項研究中,我們首先驗證了潛在組蛋白甲基轉移酶抑制劑在多種癌細胞株中的生長抑制作用。在 14 種候選藥物中,DHT-07343和DHT-07171 兩種化合物對細胞增殖表現出最明顯的抑制效果,在大腸癌細胞株中尤為顯著。我們接著證實這兩種化合物可藉由調節幾種組蛋白甲基轉移酶來抑制酶的活性。此外,也證明了 DHT-07343和DHT-07171對癌細胞轉移的抑制行為,這可能降低腫瘤發展中的癌細胞轉移和侵襲風險。同時,我們通過西方墨點法測定與細胞凋亡、細胞週期和上皮間質轉化(EMT)有關不同訊息傳遞途徑的生物標記,以分析藥物於生物學功能之作用。另外,我們的化合物在與化學治療藥品的合併使用下,無論於常規或耐藥細胞株中皆能表現增強的抗增殖效果。而為了驗證藥物的安全性,我們將DHT-07343和DHT-07171使用在人類正常細胞株和斑馬魚胚胎毒性試驗中。
總結來說,我們的研究顯示組蛋白甲基轉移酶抑制劑DHT-07343和DHT-07171 可以通過抑制細胞增殖、轉移和耐藥性來阻遏癌症發展,為大腸癌治療提供了一種有前景的策略。
