Abstract
許多內部和外部刺激可能導致肝臟發炎和膠原纖維堆積,影響肝功能並增加罹患肝硬化和肝癌等疾病的風險。米糠是稻米加工的主要副產品,含有多種對肝臟有益的營養成分。在本研究中,我們研究了米糠對四氯化碳 (CCl4) 誘導小鼠肝纖維化的影響。我們將小鼠分為四組,分別餵食含有10%米糠的飲食或標準飲食,並注射20% CCl4以誘導肝纖維化。結果顯示食用含有米糠的飲食能有效減緩 CCl4 誘導的肝損傷、膠原纖維積聚,以及與纖維化相關的基因表達,包括 α-平滑肌肌動蛋白 (α-SMA)、膠原蛋白 1a2 (COL1A2) 和轉化生長因子-β (TGF-β)。此外,食用米糠提高了第二階段解毒和抗氧化基因的表達,包括 Gsta3、Gstp1、过氧化氢酶、SOD1、SOD2 和 SOD3。米糠中的主要生物活性物質γ-穀維素降低了肝星狀細胞對TGF-β1誘導的α-SMA、COL1A2和磷酸化smad2/3表現的敏感性。並且,γ-穀維素 還增加了 HepG2 细胞中第二階段解毒基因 GSTM、SULT1A1 和 SULT1A3 的表達。我們的研究顯示,含有米糠的飲食對改善肝纖維化具有潛在的益處,主要通過增強肝臟抗氧化和解毒活性。並且,米糠中的主要生物活性物質γ-穀維素能夠抑制肝星狀細胞活化以及增加肝細胞的解毒功能。
