Abstract
關節軟骨是一種透明的組織結構,覆蓋在骨關節的表面,提供光滑的低摩擦接觸表面。它主要由分佈稀疏的特化軟骨細胞和大量的細胞外基質(ECM)組成。 與人體中的大多數組織不同,關節軟骨缺乏淋巴組織和血管,由於損傷後血液供應不足,限制了其自我修復的能力。關節軟骨的累積損傷可導致不可逆的骨軟骨破壞和退行性關節炎等病症。目前可用於關節軟骨修復的多種治療方法包括微骨折、自體軟骨細胞移植和自體骨軟骨移植。然而,這些方法中的每一種都有明顯的局限性和缺點。例如,微骨折會導致軟骨纖維化,自體軟骨細胞移植可能導致軟骨整合不良,而自體軟骨移植會導致供體部位出現並發症,並且骨採集來源有限。為了應對這些挑戰,我們開發了一種無藥物、微創的工程軟骨修復方法,使用由電漿交聯的生物粘合劑凝膠。對於在治療抑鬱症、焦慮症、過敏和頭痛方面具有生物學和臨床功能的酪胺(TYR),在化學上與藻酸鹽(ALG)主鏈進行交聯,形成 ALG-TYR 前體。隨後,經過電漿處理五分鐘可增強水凝膠的機械性能並賦予水凝膠狀表型穩定性。開發的ALG-TYR+P/5min 水凝膠系統具有生物相容性,可有效抑制與類風濕性關節炎 (RA) 相關的炎症反應。此外,它還具有調節受損區域免疫反應的能力。這種仿生和自我修復水凝膠提供了一種安全有效的治療選擇,並降低了副作用,從而加強了對RA的治療。從這項研究中獲得的結果為改進 RA 治療方法提供了有希望的前景。
