Abstract
長期處於高血糖情況,生物體會因多元醇途徑(polyol pathway)而產生甲基乙二醛(methylglyoxal;MGO)等糖毒素,進而造成細胞的凋亡。由於粒線體的形態與細胞凋亡的關係密切,本研究探討MGO是否造成粒線體之形態改變。本論文以穩定表達轉染粒線體enhanced yellow fluorescent protein (EYFP)之HEK-293細胞進行實驗,發現以MGO (300 μM) 處理,會伴隨粒線體的分裂增加。但對與粒線體分裂相關的基因,如Drp 1、hFis 1、mitofusin 1、mitofusin 2等進行蛋白質與核醣核酸表現的分析則沒有發現差異,而只有活性氧自由基(reactive oxygen species;ROS)的量增加。本論文再以臨床上用以治療第二型糖尿病的藥物metformin處理細胞,發現其可以減少粒線體的分裂及ROS的產生。利用AMP-activated protein kinase (AMPK)活化劑AICAR或metformin處理細胞,發現metformin和AICAR都可以增加AMPK的磷酸化,。本研究顯示methylglyoxal可影響粒線體型態改變,而此一作用可被胰島素增敏劑,metformin透過AMPK的活化而逆轉。
