Abstract
血管平滑肌細胞的不正常增生和遷移,會造成血管內膜的異常增厚,
在很多心血管疾病,例如動脈粥狀硬化及血管再窄化的病程中都扮演了相當重要的角色。本篇論文在探討ketamine,一種快速作用、非巴比妥類藥物的解離性(dissociative)麻醉劑,抑制大鼠血管平滑肌細胞增生的分子機轉。本篇論文發現ketamine對於血小板衍生生長因子(platelet-derived growth factor-BB, PDGF-BB)誘導的大鼠血管平滑肌細胞增生具有抑制的效果。進一步探討ketamine對於PDGF-BB訊息傳遞路徑的調控,發現ketamine能抑制PDGF-BB所誘導的磷脂醯肌醇-3激酶(PI3K)與蛋白激酶(Akt)及胞外訊息調控激酶1/2(ERK1/2)的磷酸化,而蛋白去磷酸酶2A(PP2A)的抑制劑okadaic acid能回復ketamine抑制PDGF-BB所誘導的Akt及ERK1/2磷酸化。此外本篇論文也發現ketamine能抑制PP2A在Tyr307的磷酸化和Leu309的去甲基化。從實驗結果推論,ketamine 可能是透過抑制PP2A的蛋白修飾作用,特別是在Tyr307磷酸化和Leu309去甲基化,使得PP2A活性增加,並進
一步造成Akt和ERK1/2的去磷酸化,最終抑制 PDGF-BB 所誘導大鼠血管平滑肌細胞所誘導大鼠血管平滑肌細胞的增生作用。
