Abstract
乳癌為全球女性最常見的癌症之一,其致死率居高不下,主要原因為乳癌容易轉移至其他器官,導致病人的病情惡化而死亡。儘管過去十年,手術和化學治療技術日益增進,乳癌的治療及預防腫瘤轉移仍然遇到許多瓶頸。約有3-15%的乳癌病人會因手術受到細菌的感染,當病人受到細菌的感染時,則會促進腫瘤細胞之轉移。因此,利用天然物有效抑制乳癌細胞的轉移,有助於臨床乳癌治療。實驗結果發現,LPS確實具有促進乳癌細胞株MCF-7和MDA-MB-231的移行能力,細胞形態也受到改變,同處理hispolon後,其乳癌細胞移行受到抑制,並且抑制 LPS誘導的細胞形態改變。分析間質標的 (Mesenchymal markers) Fibronectin和Vimentin蛋白質及mRNA,其表現量受到抑制,進而抑制下游MMP-2、MMP-9和u-PA (PLAU) mRNA和其酵素活性,反觀,上皮標的 (Epithelial markers) E-cadherin和β-catenin的蛋白質及mRNA的表現量增加。Hispolon抑制上皮間質轉型過程中,其透過抑制轉錄因子NF-κB和AP-1的活性。此些結果顯示,hispolon或許具有潛力作為未來治療乳癌病人轉移的藥物之一。
