Abstract
DnaJA3 是屬於 Dna J family 的成員之一,在過去的研究指出,可能扮演腫瘤抑制蛋白 (tumor suppressor) 的角色。在人類基因的研究中,發現有一個 Tid56 的同源蛋白稱為 hTid1 (human tumourous imaginal disc),是一種 co-chaperone protein。hTid1 主要具有兩種不同剪接型式的 variant,分別為 Long form 及 Short form。利用 hTid1 序列搜尋斑馬魚基因庫,發現斑馬魚有兩個與 hTid1 類似的基因,分別為 DnaJA3a 與 DnaJA3b。本論文主要的研究目的為探討 DnaJA3a 與 DnaJA3b 在斑馬魚發育時期功能之差異性。利用 RT-PCR 與完整胚體雜交技術 (whole-mount in situ hybridization) 分析 DnaJA3a 與 DnaJA3b 在斑馬魚胚胎發育過程中表達的時間和位置,結果顯示,DnaJA3a 在胚胎發育 12~120 hpf 各個時期都較為明顯,而 DnaJA3b 在受精後 72 小時表現較明顯,其他時期則不明顯;而在成熟個體組織表現分析顯示,DnaJA3a mRNA 會全身性的表現,而 DnaJA3b 則在腦部和肌肉會顯著的表現。另外利用轉殖斑馬魚的技術,分別構築 wild type (wt) 和 DnaJ 區域變異 (mutant) 的 DnaJA3a 與 DnaJA3b 的表現質體,利用顯微注射 (microinjection) 方式,將不同形式的表現質體注射入斑馬魚胚胎,藉此來探討不同形式 DnaJA3a 與 DnaJA3b 基因表現對於斑馬魚胚胎發育的影響。結果顯示,過量表現正常 DnaJA3a 會造成胚胎的突變與死亡,而過量表現正常 DnaJA3b 的部分則不會影響斑馬魚胚胎早期的發育。過量表現 DnaJA3b DnaJ 區域突變的質體,則會造成胚胎體節發育異常,而過量表現 DnaJA3a DnaJ 區域突變的質體,不影響斑馬魚胚胎外觀。本論文將藉由過量表現不同形式的 DnaJA3a 與 DnaJA3b 蛋白來探討這兩種 hTid1 相似的基因,對於斑馬魚胚胎發育和生長的影響。
