Abstract
胰臟癌 (Pancreatic cancer) 的死亡率在全球癌症死亡比例中居高不下,其中胰臟癌最常見的類型就是胰腺導管癌 (Pancreatic ductal adenocarcinoma, PDAC) PDAC 因為早期症狀不明確,也缺乏特定的生物標誌物,導致大約有 80% 以上的患者被診斷出 PDAC 時已經是晚期或有轉移的症狀發生。常見的第一線化療藥物為吉西他濱 (Gemcitabine) PDAC 患者易對此藥物產生抗藥性 ,進而導致對 PDAC 患者治療上的有效性降低。 本研究在檸檬苦素 (Limonin) 中發現,此純化物可有效抑制 PDAC 及 PDAC /GR 細胞生長與細胞爬行之能力。 除此之外, 檸檬苦素可有效抑制人類 PDAC 細胞株 PANC-1 及對吉西他濱藥物有抗藥性的 PANC-1/GR 細胞其癌症幹細胞標誌之 mRNA 表現。 在 NGS 分析中,發現檸檬苦素主要會透過下調糖解路徑,以及影響細胞週期 G2 至 M 期表 現,來延緩 PDAC 病程進展 。 總結來說,透過先前研究以及本篇研究結果,因此我們推論檸檬苦素可能會透過影響癌症幹細胞標誌 ALDH1A1 之 mRNA 表現,進一步下調糖解路徑, 來延緩 PDAC 病程進展, 這也顯示檸檬苦素可能具有成為胰臟癌藥物的潛力,但相關分子機制的部分還需要更多的研究做進一步的探討 。
