Abstract
鈷 (cobalt)為環境汙染物,並被國際癌症研究署 (International Agency for Research on Cancer, IARC)列為Group II B的致癌物質,其可造成多種肺部疾病且導致肺部上皮細胞纖維化的發生,而細胞凋亡則是在纖維化發生前之重要步驟,在許多細胞實驗中發現,氯化鈷 (cobalt chloride, CoCl2)可造成細胞凋亡,其中包括HIF-1a、P53、ROS大量表現及粒線體膜電位去極化等等,而其凋亡機制仍有待探討。本論文利用人類上皮肺癌細胞株 (non-small cell lung cancer, H460)進行實驗,企圖探討氯化鈷對H460所造成的細胞毒性及凋亡機制。實驗結果發現,氯化鈷對H460造成time- and dose-response且caspase-dependent之細胞凋亡,其中,粒線體膜電位沒有去極化現象,細胞內的H2O2顯著上升但NAC無法降低凋亡的比例。另外,氯化鈷可誘導細胞內P53及HIF-1a大量表現、YY1表現量降低及DAPK活化。FK506—calcineurin inhibitor,可抑制DAPK活化及P53表現,對細胞凋亡亦有顯著的抑制效果。推測氯化鈷經由誘導P53大量表現及DAPK活化而造成細胞凋亡。 關鍵字: 鈷,氯化鈷,calcineurin,caspase,DAPK,FK506,HIF-1a,P53,ROS,YY1
