Abstract
目前神經膠質瘤與慢性骨髓性白血病的治療仍急需改善。已知癌細胞可透過增加RNA和蛋白質的核質運輸,進而促進腫瘤生長以及迴避細胞凋亡的機制。RNA與蛋白質從細胞核釋放至細胞質的過程中必須透過RanGTPase activating protein 1 (RanGAP1)協助,因此假設抑制RanGAP1表現或許可作為改善上述兩種腫瘤治療的新策略。冬凌草甲素(oridonin)是傳統中草藥冬凌草所分離出的天然雙萜類化合物,本實驗室之前的研究已證明oridonin會減少神經膠質瘤細胞中SUMOylated RanGAP1的蛋白表現。在本研究中,我們證實了oridonin透過抑制RanGAP1表現會導致RanGTP的核堆積進而阻斷RNA輸出,最終導致U87MG神經膠質瘤細胞凋亡。此外本研究亦透過K562慢性血癌細胞證實,利用shRNA與microRNA抑制RanGAP1表現可阻斷BCR-ABL致癌蛋白的核輸出,進而活化P73蛋白及Bax蛋白表現並加強藥物的毒殺作用。綜合前述結果,本論文研究證實抑制RanGAP1表現可阻斷細胞核內RNA與蛋白質輸出,故以冬凌草甲素或microRNA抑制RanGAP1表現,進而阻斷RNA和蛋白質的核質運輸並影響腫瘤細胞之命運,或許是一個新穎有效的治療策略。
