Abstract
由於先前文獻利用DNA微陣列(Microarray analysis)偵測乳癌細胞株中相關基因的表現,發現Prim1在乳癌細胞株中大量表現;先前實驗室已探討Prim1與乳癌之間的關聯性,並未探討在癌幹細胞中與Prim1的關聯。在254個乳癌病患檢體中觀察到Prim1在乳癌病患的腫瘤組織中表現量高於正常組織,且在預後狀況以及腫瘤大小有顯著的差異,在細胞株當中證實將Prim1抑制其表現會影響細胞週期及細胞移行的能力。
本研究初步分析Prim1在13株乳癌細胞株進行RT-PCR及西方點墨法分析,發現Prim1在乳癌細胞株中mRNNA及蛋白質表現相較正常乳腺細胞有高度表現,接著在13株乳癌細胞株當進行RT-PCR分析幹細胞的特性,並分析側群細胞(Side Population, SP)的比例,及分析細胞的CD44+/CD24-的表現建立乳癌幹細胞的分析方式。之後在穩定表現siPrim1的細胞中分析側群細胞與CD44/CD24的表現,發現在側群細胞或表現CD44+/CD24-的細胞的比例皆有下降,並觀察野生型MCF-7分選出CD44+/CD24-的細胞中Prim1的表現高於CD44+/CD24+的細胞,並利用RT-PCR分析發現Prim1與幹細胞特性基因(Sox2、Oct4、β-catenin及Notch1)有相關性。在siPrim1分選出CD44+/CD24-的細胞表現的腫瘤球體(Tumorsphere)明顯小於由野生型MCF7分選出CD44+/CD24-的細胞。接著利用天然萃取物Inotilone可抑制Prim1的表現且抑制野生型MCF7細胞的生長與側群細胞的比例;因此本篇研究推論Prim1與癌幹細胞可能具有相關性,並期望未來能夠使用天然萃取物降低癌幹細胞的比例以增加臨床治療的效果。
