Abstract
氣喘為一種肺部慢性發炎疾病,約有10% 的人口深受其擾。Thrombin 為 serine 蛋白酶,是已知凝血因子,可從受傷血管釋放出來,且在肺部的發炎扮演著非常重要的角色。IL-8/CXCL8 是一種嗜中性白血球趨化因子,在氣喘病人中參與調節發炎細胞的移動。許多研究顯示,CAAT/enhancer binding protein-β (c/EBPβ) 在調控呼吸道發炎基因的表現扮演著重要的角色。我們先前研究指出,c/EBPβ 參與 thrombin 誘導肺部上皮細胞 IL-8/CXCL8 表現。因此,本論文將進一步探討p300依賴c/EBPβ乙醯化及c/EBPβ 調控 IKKβ/IκBa/NF-κB 訊息路徑參與 thrombin 誘導肺部上皮細胞 IL-8/CXCL8 的表現。在本研究中我們發現 thrombin 誘導 IL-8/CXCL8-luciferase 活化可被 p300 siRNA 抑制。另外,thrombin 誘導 p300 與 c/EBPβ 複合物的形成及依時間相關增加 c/EBPβ 乙醯化,而此增加的情形也可被 p300 siRNA所抑制。我們也發現 thrombin 誘導 κB-luciferase 活化及 IκBa 磷酸化的增加受到 c/EBPβ siRNA 抑制。再者,thrombin 增加人類肺部上皮細胞 IKKβ蛋白表現及磷酸化,但卻不影響 p65蛋白表現。最後我們發現 thrombin 誘導 p65 及 c/EBPβ 複合物形成且誘導 c/EBPβ 與 p65 及 p300 形成複合體,結合至 IL-8/CXCL8 promoter 上,增加 IL-8/CXCL8 轉錄的作用。綜合以上結果,我們發現 p300依賴 c/EBPβ 乙醯化和 NF-κB 活性的調控促進thrombin誘導人類肺部上皮細胞 IL-8/CXCL8 表現。本研究結果將有助於釐清 c/EBPβ 訊息路徑參與 thrombin 誘導人類肺部上皮細胞 IL-8/CXCL8 表現。
