Abstract
PFCs為一種人工製作化學物,分為全氟辛磺酸(perfluorooctane-sulfonate, PFOS)、全氟己烷磺酸 (perfluorohexyl-sulfonate, PFHxS)、全氟辛酸 (perfluorooctanoic acid, PFOA) 和全氟壬酸 (perfluorononanoic-acid, PFNA) 四種。這些化學物在我們日常生活中多在使用,而且對人體上有潛在的危害。近年研究指出在美國多於百分之九十之人血液中偵測到PFCs,累積於肝臟及腎臟。近年來PFCs中的化學物PFOS和PFOA對人體健康和環境的影響備受關注,先前文獻有提到PFOS為PFCs中毒性最強,並且與過氧化物酶體增殖物激活受體PPARs(peroxisome proliferator activated receptor)中的PPARγ有關連性,PPARγ透過PFOS的作用下增加腎臟的損傷;但另外文獻指出PFOS影響肺臟或肝臟的損傷與PPARγ無關,故到目前為止PFOS和PPARγ之間的關係尚未驗證。其他文獻提到PFOS引發細胞的型態改變,並增加遷移能力。本篇實驗是以針對此化學物PFOS對腎小管細胞生理上的作用,結果證明腎小管細胞在PFOS作用下,促進Sirt1去乙醯化PPARγ和p53,導致細胞受氧化壓力、細胞凋亡,以及細胞走向表皮間質轉型(Epithelial-Mesenchymal Transition, EMT),而這些反應可被L-carnitine抑制。
