Abstract
背景:近年針對肺癌之藥物進步如免疫抑制劑,所帶來的益處往往只在一小部分的患者中顯現,研究指出免疫微環境會影響化療反應。相較於單一生物標記,結合多種免疫組織概況可獲得更好癌症預測。方法:本研究搜集TCGA 資料庫LUAD 患者(n=513)基因表達及臨床資料,以及GEO 非小細胞肺癌患者(n=42)單細胞資料,透過Bisque 細胞分解工具拆解患者之細胞比例,並整合多篇文獻之免疫特徵建立新免疫圖,運用ssGSEA 免疫分數繪製免疫圖譜,另以Lasso-Cox model 建立PPS 23個與PFI 相關基因,彙整以上特徵進行K-means 分群,依照存活分為C1 組及C2 組,進行化療病患免疫特徵分析及預測模型建立。結果:C1 組相較C2 組具備顯著存活優勢,且帶有之免疫細胞比例及免疫評分均優於C2 組。結論:本研究證實細胞比例的拆解、免疫特徵及PPS 對於化療反應有關聯性。
