Abstract
阿茲海默症是一種漸進性的神經退化疾病,其診斷相當依賴臨床人員的經驗,對於及早治療阿茲海默症是不足的,故希望能從病人血清中尋找出有顯著差異的蛋白生物標誌,幫助疾病之診斷、追蹤和治療。研究發現神經退化疾病”亨丁頓舞蹈症(Huntington Disease; HD)”的關聯蛋白(Huntingtin associated protein; HAP)1不但與HD有關,也參與阿茲海默症蛋白-類澱粉前驅蛋白(amyloid precursor protein; APP)的運輸調節。HAP1的相關蛋白在阿茲海默症病人血清中是否有改變,以及他們在神經退化疾病上所扮演的角色,是本研究中所要探討的。我們取得健康對照組與阿茲海默症組的血清,分析受試者的臨床數據後,在基礎生化檢驗項目中沒有明顯差異並且與疾病嚴重程度無顯著相關,再使用西方墨點法確認血清中HAP1相關蛋白的含量,發現阿貝爾森輔助者融合位點蛋白(AHI-1; Jouberin)、動力蛋白輕中鏈(DYNC1LI2)以及驅動蛋白輕鏈 (KLC2) 在阿茲海默患者血清中顯著降低。酵素結合免疫吸附分析(ELISA)也有一致的結果。我們將更進一步建立可供驗證的細胞模型,以檢測當這些蛋白過多或過少時對細胞產生何種影響。
