Abstract
基質金屬蛋白酶(Matrix metalloproteinases, MMPs)不管在維持正常生理功能或病理現象上均扮演重要角色;MMPs參與正常皮膚功能,如傷口癒合和老化,同時也和胚胎發育、生殖功能,以及發炎反應相關,並與癌症入侵以及轉移,和關節炎發生具關連性。我們所感興趣的部份是皮膚發炎反應和MMP-1,-2及-9活性的相關性,有研究指出在一些皮膚炎的病例中檢測出較高的MMP-1及-2活性,而在一些難以癒合的皮膚傷口中也發現MMP-9大量累積,在本篇研究我們測試數種常見中藥白芷 (Angelica dahurica)、川芎 (Ligusticum chuanxiong)、當歸 (Angelica sinensis)、黃芩 (Scutellaria baicalensis)、黃耆 (Astragalus membranaceus)、覆盆子 (Rubus chingii)、防風 (Ledebouriella divaricata)、山藥 (Dioscorea opposita)、丹參 (Salvia miltiorrhiza)、牡丹皮 (Paeonia suffruticosa)和甘草 (Glycyrrhiza uralensis)尋找能抑制MMP-1,-2及-9活性的藥物,以期在治療皮膚炎上能加以利用。我們以腫瘤壞死因子-α (Tumor Necrosis Factor-α, TNF-α)作為刺激劑,刺激人類纖維母細胞WS1大量表現MMP-1及-9,MMP-2的表現並不受TNF-α的影響。在測試的11種中藥萃取物皆能有意義的降低由TNF-α引發的MMP-1之活性;而其中牡丹皮、防風、山藥、覆盆子、甘草、丹參則是會抑制MMP-9之活性,而MMP-2活性則會有意義的受防風和覆盆子抑制。TNFα則不會影響Tissue Inhibitor of MMP-1 (TIMP-1)之分泌。在對於Interleukin-8 (IL-8)表現量的測試上則發現覆盆子可以大幅降低由TNF-α引發的IL-8表現,而IL-8在過去的研究中被認為會造成MMP-9的分泌。這些結果顯示,牡丹皮、山藥、防風、覆盆子、甘草及丹參具有抑制MMP-1及-9活性的成份,在皮膚炎的治療或是加速皮膚傷口癒合上有著相當的潛力。
