Abstract
雖然人工製備的多孔質生醫材成型物被廣泛用於組織工程中,但生物的高次結構及孔洞型態卻是人工難以模仿的,因此利用異體或異種動物的去細胞之組織,是近期被發展的技術之一;然而動物組織的複雜度高,去細胞效率也不易控制而影響了它的發展;相對地,植物組織的均一性遠高於動物組織但基質的生理活性則低於動物組織,所以本研究欲將去細胞的植物組織細胞間質經表面化學改質賦予生理活性,達到重建動物組織的目的。首先,我們收集不同型態包括根、莖、葉、果實及花等植物部份包括蘋果、蘆薈,菠菜,青龍葉(iris virginica),著我(Iris japonica)等,經界面活性劑處理後去除植物細胞,在乾燥完成後以掃瞄式電子顯微鏡進行觀察、孔洞測量及切片染色以確定去細胞後的結構;樣本結構的選定主要收集各種具不同大小孔洞且不同配向構型的植物組織細胞間質,測試其強力等物理性質,其中蘋果支架,具軟組織或表皮組織應用的潛力。菠菜葉具有葉脈構造,可以做為血管、腺體等應用。青龍葉、著我為平行葉脈,可作為的如配向結構的肌腱之支架。我們以NIH-3T3進行生物相容性、細胞貼附及細胞增生等生物測試,我們將擇優的植物組織細胞間質接枝聚肤胺酸、幾丁聚醣,進行表面化學改質以賦予較高的生理活性,再以不同的初代細胞培養證明其可用性;希望不久的將來用應用於臨床醫療之上。
