Abstract
非酒精性脂肪肝疾病(nonalcoholic fatty liver disease, NAFLD)是停經後婦女之常見疾病,特徵為其特徵為停經狀態之下,後因雌激素缺乏所而導致的脂質代謝紊亂異常以及肝臟脂肪累積的堆積。過去研究結果已知顯示,omega-3多不飽和脂肪酸(polyunsaturated fatty acid, PUFA)有助於調節脂質代謝異常、降低非酒精性脂肪肝疾病NAFLD的發生率。卵巢切除動物模型式目前已廣泛用於模擬婦女停經後狀態,我們先前的研究發現,富含omega-3 PUFA、(EPA及DHA)的魚油飲食使得卵巢切除大鼠之有較嚴重的脂肪肝的情況加重惡化,而富含α-亞麻酸的紫蘇油飲食組別則有之脂肪肝卻顯著減減輕少的情況。因此,本研究的主旨在於闡明探討魚油和紫蘇油(兩者富含omega-3 PUFA)相對於玉米油(富含omega-6 PUFA)等飲食,在卵巢切除大鼠中所調控影響的的分子機制有何不同。我們利用定量蛋白質體學的方式技術分析了卵巢切除大鼠的肝臟組織,相較於以玉米油之標準組別為基準,在魚油介入和紫蘇油介入的組別分別檢測鑑定到244和196個具有差異表現量明顯不同的蛋白質。我們透過PANTHER和IPA等軟體分析這些表現量不同的蛋白質功能,結果顯示在魚油介入的組別中,揭示了這些蛋白質與脂肪酸代謝、過氧化小體組成、免疫細胞的趨化及募集聚集等細胞功能皆為過程的上調正調控,以及而免疫反應的下調相關則是負調控; ; 在紫蘇油組中,受調控的異常表現量之蛋白質則主要和較上游的脂肪酸代謝的上游過程、脂質運送、急性炎症反應的下調負調控、以及對於有毒物質的反應有關。在硫代巴比妥酸脂質過氧化反應物質分析指標(TBARS analysis)分析結果中,我們證實顯示顯示魚油組的脂質過氧化程度上升,而骨髓過氧化酵素髓過氧化物酶活性測定(MPO activity assay)印證了魚油組中證實了發炎情況的下降的現象。以上我們本的研究結果都顯示,了魚油和紫蘇油飲食對於卵巢切除大鼠具有截然不同的分子作用機制。
透過這根據此個研究,我們可提供臨床上停經後婦女選擇omega-3 PUFA未來不只能在臨床上給予不同需求的病人提供omega-3 PUFA的供給選擇油脂來源的參考依據。,也可能針對不同病程階段的病人做飲食結構的調整。
