Abstract
替代性剪接是高等真核細胞中增加基因表現多樣性的重要機制,能將同一基因中的外顯子以不同的組合方式,轉譯出含有不同胺基酸序列的蛋白質亞型。這種機制在不同的組織中或發育時期受到嚴密的調控,錯誤的替代性剪接機制造成多種疾病的發生,而目前替代性剪接對脂肪細胞的功能仍不清楚。脂肪組織在哺乳類動物中調節生物體內能量的平衡,能量平衡的失調導致代謝性疾病的產生。目前已知在哺乳動物中存在白色脂肪組織與棕色脂肪組織,棕色脂肪組織可以產熱來消耗能量,白色脂肪組織則可以儲存能量。我們利用全轉錄組定序法分析不同時期野生型與RBM4a剔除之小鼠棕色脂肪組織,發現Mitogen-activated protein kinase kinase kinase kinase 4 (MAP4K4) 基因的表現量或不同轉錄子的表現量,在不同時期皆會受到影響。過去研究指出,RBM4可調控棕色脂肪細胞中相關基因的替代性剪接,以及MAP4K4會造成白色脂肪細胞產生胰島素抗性。本篇論文的研究目的即是探討在棕色脂肪細胞生成時,RBM4如何調控MAP4K4基因的替代性剪接,以及MAP4K4蛋白亞型對棕色脂肪細胞生成的影響。經由結果分析,我們發現MAP4K4蛋白亞型有助於棕色脂肪細胞的分化或發育,以及不同的MAP4K4蛋白亞型在棕色脂肪細胞分化的不同時期,有其特定的功能表現。除此之外,RBM4在棕色脂肪細胞生成時會調控MAP4K4基因的替代性剪接。因此,我們推論MAP4K4會受到RBM4的調控參與棕色脂肪細胞的分化或發育。
