Abstract
幹細胞在組織工程的發展上已日趨重要,其中牙髓幹細胞可以經由非侵入式的方式取得,相較於胚胎幹細胞及骨髓幹細胞,取得方式較為單純,爭議性也較少。近年來,技術上的進步使得牙髓幹細胞可成功的被應用於牙周方面的疾病和下顎骨的再生,也因幹細胞的自我再生和多重分化能力而使其具有更多方面應用的潛力。透明質酸為一種天然的生物性聚合物,在脊椎動物甚至細菌內都有著重要的生理功能,目前被廣泛應用於醫療及美容產業。透明質酸不僅為組織工程中一個重要的材料,且有文獻指出其與特定細胞甚至幹細胞的生理活動有關,因此本實驗欲探討透明質酸對牙髓幹細胞的影響。
本實驗使用細胞外生法取得初代的牙髓幹細胞,並以表面特徵蛋白鑑定以及分化能力測試來確認其保有牙髓幹細胞的特性。在透明質酸影響細胞生長的實驗中,在培養液內加入透明質酸,再以MTT法測試細胞的增生情況。另外也以透明質酸之主要受體CD44的抗體來阻隔兩者的接合位置,以確認透明質酸的影響是透過與CD44的接合。MAPK pathway與細胞的生長有關,又分為ERK、JNK、p38 MAPK三條路徑,本實驗使用這三條路徑的抑制劑PD98059、SP600125、SB203580,來檢測透明質酸是透過哪些路徑來影響細胞的生長。最後也在牙髓幹細胞誘導分化的實驗中添加抑制劑,來探討是否透明質酸能影響幹細胞的分化能力。
實驗結果發現初代培養的牙髓幹細胞能夠表現幹細胞的特徵蛋白,也能成功的進行骨分化、軟骨分化及脂肪分化。根據MTT實驗的結果可發現,在無血清培養的狀況下,添加1 mg/ml的透明質酸於培養液內可以促進牙髓幹細胞的增生,在細胞培養第三天之後可以看到顯著的差異,且此影響是透過透明質酸與CD44的結合。MAPK pathway的實驗結果發現透明質酸對於牙髓幹細胞的影響與JNK及p38 MAPK兩條路徑相關,與ERK路徑較無關係。另外在誘導牙髓幹細胞分化約三週後染色,結果顯示1 mg/ml透明質酸對牙髓幹細胞的分化能力並無影響。因此我們可以總結,1 mg/ml的透明質酸能夠在不影響牙髓幹細胞分化能力的情況下與CD44結合,並影響JNK及p38 MAPK 兩條路徑,從而促進細胞的增生。
