Abstract
中文摘要

神經膠質瘤在成年人的中樞神經系統中,是最常見的惡性腦腫瘤。其病理特徵為癌細胞的過度增殖、細胞遷移以及瀰漫至周圍腦組織,造成抗腫瘤的免疫監督系統失去功能,皆為關鍵的生物特點,有助於鑒定腦膠質瘤惡性表型。儘管現在的合併療法(combined therapies)不斷進步,包括手術、放射治療以及化學療法,但患者的預後並沒有獲得良好的改善。轉化生長因子-β(Transforming growth factor-beta;TGF-β)扮演關鍵的路徑在調控細胞外基質基因的表達,當它被過量誘導時,會參與調控細胞遷移過程。厚朴酚(Magnolol;MAG),一個從厚朴純化出來的化合物,這是目前用於研究抑制人腦膠質瘤細胞增殖。在這裡我們研究,厚朴可以抑制TGF –β訊息傳遞路徑所誘導的C6神經膠質瘤細胞的遷移行為及其相關機制。我們發現,轉化生長因子-β誘導神經膠質瘤的細胞遷移,與處理的時間呈相關性。轉化生長因子-β造成的前遷移反應 (pro-migratiry effects),明顯受到厚朴酚所抑制。厚朴可降低TGF –β所調控的Smad蛋白磷酸化,並且與時間呈現正相關。我們也證明厚朴可降低轉化生長因子-βⅠ型受體 (TGF-β type I receptor)。這些結果顯示,厚朴可當成一個潛在性抑制惡性神經膠質瘤細胞遷移行為的研究標的。
