Abstract
中樞神經系統損傷後會伴隨有神經膠變性 (reactive gliosis) 現象,該現象包含神經膠細胞的肥大與增生。在中樞神經系統及腦血管損傷中,thrombin會誘導凝血反應、神經膠變性與免疫作用。此外,結締組織生長因子 (connective tissue growth factor, CTGF) 亦被認為在血管新生與組織修復中扮演重要角色,在神經系統中亦與gliosis有關。然而thrombin刺激CTGF表現詳細的分子機轉目前還不清楚,在本論文中我們將探討thrombin調控CTGF表現的訊息傳遞分子機制。Thrombin會以濃度相關性與時間相關性增加CTGF表現及釋放。使用protease activated receptors (PARs) 促效劑發現僅有PAR-1促效劑有類似thrombin刺激CTGF表現,而使用PAR-3及PAR-4促效劑則是沒有作用;另一方面,給予PAR-1拮抗劑可降低thrombin所誘導CTGF表現。使用dominant negative of c-Src (c-Src DN) 依濃度相關地抑制thrombin所誘導CTGF表現,且thrombin會依時間相關地增加c-Src之Tyr416磷酸化;此外,使用SP600125 (JNK 抑制劑) 與JNK1 DN及JNK2 DN也會抑制thrombin所誘導CTGF蛋白的表現,我們也發現thrombin可依時間相關性增加JNK之Thr183 / Tyr185磷酸化。Curcumin (AP-1 抑制劑) 可抑制thrombin誘導CTGF表現,且thrombin可增加AP-1-luciferase活性。而給予c-Src DN、JNK1 DN或JNK2 DN可以抑制AP-1-luciferase活性。此外,本論文也證實thrombin與CTGF均會誘發C6神經膠瘤細胞的增生,且使用CTGF siRNA會降低thrombin所誘導的細胞增生。綜合以上結果發現thrombin可經由PAR1誘導c-Src / JNK / AP-1訊息傳遞路徑來誘導CTGF表現,並且CTGF可能參與thrombin所誘導C6神經膠細胞之增生。
