Abstract
能量代謝重塑是腫瘤的標誌生理過程之一,為了探討隱丹參酮(Cryptotanshinone, CTS) 調控乳腺癌細胞能量代謝的分子機制,本研究應用定量蛋白質組學技術研究隱丹參酮對MDA-MB-231細胞蛋白質組的影響,結果顯示隱丹參酮可能通過抑制MDA-MB-231細胞中電子傳遞鏈複合體的表達,實現對粒線體氧化磷酸化(Oxidative Phosphorylation, OXPHOS)的抑制,從而抑制腫瘤細胞能量代謝。進一步研究了隱丹參酮對乳腺癌細胞糖酵解和Pyruvate Kinase M2(PKM2)/β-catenin信號通路的影響,發現CTS還可以下調糖酵解相關蛋白PKM2、Lactate Dehydrogenase A(LDHA)和Hexokinase-2 (HK2)表達水準,降低PKM2和β-catenin的水準,並抑制乳腺癌細胞系的遷移和侵襲。說明隱丹參酮可以通過調節蛋白質表達,下調乳腺癌細胞糖酵解和氧化磷酸化水準,抑制細胞能量代謝。CTS還可以通過調控PKM2/β-catenin信號通路抑制MCF-7和MDA-MB-231細胞遷移,從而發揮抗乳腺癌的作用。
