Abstract
氣喘主要為過敏原所引起的呼吸道慢性發炎及氣道過度反應而導致支氣管過度收縮,氣流阻塞和呼吸不順的一種常見疾病。而氣喘最主要的特徵—呼吸道重塑 (airway remodeling),由於過敏或者發炎反應發生,使得肺受到損傷後,造成大量的胞外基質(extracellular matrix)沉積在黏膜下層、呼吸道平滑肌的增生,以及纖維母細胞的堆積而導致。第一型類胰島素生長因子 (insulin like growth factor-1,IGF-1)可刺激纖維母細胞,促進細胞增生並造成胞外基質如:膠原蛋白、彈力蛋白堆積;文獻指出,IGF-1對於氣喘所誘發的呼吸道重塑扮演很重要的角色。窖蛋白-1 (caveolin-1,cav-1),是組成細胞膜上的細微坑洞 (caveolae)的蛋白質,cav-1參與調控許多生理反應,包括:胞吞作用、鈣離子的釋放、以及生長因子受訊號傳遞等。此外,呼吸道由於平滑肌所分化的型態不同,也會造成cav-1表現量有所差異。為了要更進一步釐清IGF-1與cav-1對氣喘所造成的呼吸道重建之影響,我們利用BALB/c小鼠,以腹腔注射過敏原--卵白蛋白 (ovalbumin) 致敏,再經由氣管吸入含卵白蛋白的噴霧刺激,誘發過敏性氣喘的動物模式進行研究。實驗結果顯示,氣喘小鼠肺組織中,第一型類胰島素生長因子基因表現增加而cav-1下降,且cav-1可能與磷酸化的第一型類胰島素生長因子接受體有交互作用。
