Abstract
肺癌是一種肺部的惡性腫瘤,特徵為肺部組織中的細胞不受控制地生長。惡性腫瘤尤其是肺腺癌細胞會侵入及轉移擴散至其他肺部組織或身體的其他部分,是癌症患者治療失敗的主要原因,為近年來世界死亡率最高的癌症,而腫瘤的侵入與轉移與會作用於細胞外基質的MMP-2與MMP-9的產生有很大的關連,此外先前的研究也指出,天然食物中的抗氧化劑有良好的抗癌效果。
近年的研究顯示,天然植物萃取物成為開發治療癌症新藥的大宗
,hinokitiol是一種天然的生物活性化合物,又名檜木醇,富含於柏科植物中,其成分首先被從台灣白樺的心木中分離出來。先前的研究顯示其會抑制大鼠血管平滑肌細胞與血小板的凝集,同時也有抗發炎、抗腫瘤、抗血液凝集、抗氧化及殺菌的作用,然而hinokitiol對人類肺腺癌的抗癌作用的詳細分子機制仍然不明確。本篇論文主要探討hinokitiol促進人類肺腺癌細胞凋亡及抑制細胞轉移、氧化壓力的分子機轉。結果顯示hinokitiol會促進A549細胞凋亡以及抑制A549細胞轉移,經由MTT assay,發現單獨給予hinokitiol可抑制人類肺腺癌細胞A549的viability。再經由Western blot analysis發現hinokitiol會增加細胞凋亡路徑訊息傳遞因子p53、Bax、Cyt c、caspase-9和caspase-3的蛋白質表現量,降低腫瘤轉移相關蛋白MMP-9與MMP-2之蛋白質表現。此外經由native-PAGE analysis我們發現hinokitiol還可以提升抗氧化酵素超氧化歧化酶(superoxide dsismutase, SOD)和過氧化氫酶(catalase, CAT)的酵素活性。
綜合本篇的實驗結果我們推論hinokitiol不單經由增加p53的表現量而活化其下游訊息傳遞分子Bax、Cyt c、caspase-9及caspase-3來引導腫瘤細胞凋亡,也會透過增加其抗氧化酵素SOD與CAT之活性與抑制MMP-9和MMP-2的表現量以及清除OH•自由基來降低腫瘤的轉移及侵入作用。
