Abstract
豬流行性下痢病毒 (PEDV)屬於Coronaviridae中Alphacoronavirus,對懷孕的豬隻造成腹瀉、乳腺缺乏、生孕週期不正常等病症,且對於新生的豬隻造成急性腹瀉、嘔吐、脫水和高死亡率。在亞洲國家,像是中國、韓國、日本、台灣、菲律賓、泰國等,造成經濟上的損失。2013年4月加拿大、美國、墨西哥再次出現豬流行性下痢病毒的疫情,另外在不同的地區也出現了豬流行性下痢病毒的突變株使得疫情和經濟上的損失更為嚴重,說明了豬流行性下痢病毒能夠躲避當今疫苗,此外也證實新疫苗與治療方式開發的重要性。在本篇論文中,將由台糖研究所提供重組豬流行性下核鞘重組蛋白基因,進而製作豬流行性下痢病毒重組抗體,包括經豬流行性下痢病毒免疫過產生的雞蛋IgY,我們已經成功地得到對於PEDV核鞘重組蛋白具有特異性結合能力之多株抗體,後續利用phage display技術篩選抗體基因,成功地建立了5.3x106與4.8x106 anti-PEDV-N抗體基因庫,並成功篩選出了17株對於PEDV-N具有特異性結合能力的scFv單株抗體,其中共有14株單株抗體完成抗原決定位的測試,並且可分為辨認兩種不同抗原決定位,其中有6株anti-PEDV-N單株抗體可以進行純化,分別為anti-PEDV-N S1、S7、S9、S10、S12和L12 scFv單株抗體,可應用於未來發展豬流行性下痢病毒之快篩試劑以預防豬流行性下痢病毒的爆發。
