Abstract
本篇研究主要是要探討SCRN1在乳癌細胞中所扮演的角色。根據之前的文獻發現SCRN1在乳癌細胞中表現量比正常的乳房細胞來的高,文獻利用microarray的方法分析幾株乳癌細胞株中基因的表現量(Charafe-Jauffret, Ginestier et al. 2006),但並沒有進一步探討基因跟癌症發生的可能性為何。
在本篇論文中主要想要研究的是SCRN1在乳癌中所扮演的角色,首先利用了乳癌病人的檢體,經由Real-time PCR分析,發現SCRN1在腫瘤表現相對高於腫瘤周邊的組織,而在組織切片染色(IHC),可以在顯微鏡下觀察到SCRN1在腫瘤細胞的表現多於正常乳房上皮細胞的表現。接下來我們也在細胞實驗中想要探討SCRN1在細胞中的角色對於腫瘤是不是有直接的相關性,我們利用了siRNA送入細胞,顯示SCRN1被knock down後細胞的生長速率降低。乳癌細胞中的SCRN1的表現會被Nicotine所誘導,且發現天然藥物Inotilone和EGCG會抑制SCRN1的表現。並發現細胞過度表現SCRN1對於細胞生長有影響。
