Abstract
幹細胞來源的細胞外囊泡(EVs)被視為新一代的幹細胞治療替代選項。由於EVs具有與親代細胞相似的功能,其磷脂雙層結構亦具低免疫原性且無腫瘤化風險,已有研究證實脂肪幹細胞來源之EV(ADSC-EVs)具促進傷口癒合、抗發炎等功能。然而,細胞自然釋放的EV數量仍不足以支援商品化或臨床應用。 為解決此問題,已有研究嘗試以物理或化學刺激提升EV產量,其中光生物調節(PBM)與幹細胞EV間的關聯性尚未被廣泛探討,但PBM已知可促進ADSC活性。本研究選用830 nm雷射照射ADSC,並分析細胞活性、爬行、ROS及ATP以篩選最佳刺激條件。進一步比較ADSC照射前後所釋放EV(ADSC-EV與PBM/ADSC-EV)之數量、大小、電位與型態差異。 結果顯示,5 J/cm² PBM可顯著提升細胞活性與EV產量,PBM/ADSC-EV數量提升6.75倍,且CD63表現亦增加,但兩者在形態上無明顯差異。本研究證實PBM可有效刺激ADSC釋放EV,具潛力作為未來提升EV產量的策略。
