Abstract
本篇探討玉山薊(Cirsium kawakamii Hayata, CK)在四氯化碳誘導肝
毒性模型下,對於甲基乙二醛(Methylglyoxal, MG)與D-lactate 之影響,以
評估MG 及D-lactate 作為組織損傷指標之可行性。將50 隻
Sprague-Dawley(SD)大鼠隨機分為五組,除正常組外,其餘四組均每週兩
次腹腔注射四氯化碳:大豆油=4:6的混合物(1 mL/kg體重)誘導肝臟損傷,
並分別灌餵水飛薊素200 mg/kg/day、低濃度玉山薊25 mg/kg/day 或高濃
度玉山薊125 mg/kg/day,實驗為期八週。於第3、6 及8 週收集血清測定
其生化指數,並於第8 週收集肝組織,以切片病理染色鑑別肝損傷程度。
結果指出生化指數在第8 週時明顯增加,且與組織損傷量化結果一致。
在給與水飛薊素或玉山薊治療後,其血清生化指數及組織形態皆顯著改
善。此外,正常組與嚴重的肝損傷組相比其血清中MG 含量明顯地增加
(141.36 ± 7.86 vs. 245.09 ± 35.49 μM), 而肝組織中D-lactate 濃度也明顯上
升(8.56 ± 0.62 vs. 17.49 ± 2.00 nmol/mg ,正常組 vs. 損傷組),然而血清
D-lactate 在損傷組並無顯著性變化。在經過水飛薊素或玉山薊治療後,
血清MG 含量及肝臟中D-lactate 含量明顯降低(血清MG 含量 : 145.70 ±
9.32 and 119.55 ± 22.48 μM, 肝組織D-lactate 含量: 11.72 ± 1.87 and 9.64 ±
0.74 nmol/mg, 水飛薊素組及玉山薊高濃度組)。上述結果顯示,血清中
MG 含量及肝臟中D-lactate 含量極有潛力發展成為組織損傷之指標。
