Abstract
由於飲食西方化的普及,脂肪攝取量增加,造成肥胖及代謝疾病的盛行率居高不下。中年肥胖及代謝異常是提高晚年認知功能障礙風險的因素之一,同時也會造成腸道菌相失衡的情況。微生物-腸-腦軸線被視為是近期極具潛力之神經退化性疾病領域治療方向。而木寡糖是一種益生質,可以被腸道菌選擇性發酵,並改變腸道菌相組成,進而對宿主健康帶來有益影響。因此本研究旨在探討木寡糖介入是否能因為腸道菌相改善,進而改變高脂飲食所造成認知功能衰退及改善腦部認知功能相關蛋白質表現及脂質組成。C57BL/6 小鼠攝取正常對照或 60% 高脂飲食 9 週後,於 37 週齡時依據攝取木寡糖劑量各分成無、低、高劑量木寡糖組共 6 組。木寡糖介入 12 週後,於小鼠 49 週齡時,以行為試驗 Morris water maze (MWM) 測試空間學習及記憶能力,並採集糞便分析腸道菌相、海馬迴中與認知功能有關蛋白質表現量及脂質組成。結果顯示,小鼠在攝取木寡糖後,改善了因高脂飲食所造成的肥胖、體脂肪組成及海馬迴萎縮;在血液生化數值同樣發現空腹血糖及總膽固醇降低。而介入木寡糖不僅能改善代謝指標,同時也能提升空間學習及記憶能力,並在海馬迴中觀察到類澱粉斑塊堆積減少、腦源性神經營養因子增加,以及神經發炎的情況降低;腦部脂質組成也在木寡糖介入後,改變了高脂飲食所造成的 glycerolipids 代謝路徑變化。從腸道菌相分析發現攝取木寡糖後,能調節免疫力之 Faecalibacterium增加,綜合上述結果,推論木寡糖可能透過增加腸道中好菌,以影響腦部中脂質組成,進而改善小鼠腦部與認知功能障礙有關的病理指標,以延緩其空間學習及記憶能力衰退的情況。
