Abstract
蛹蟲草 (Cordyceps militaris) 是蟲草屬 (Cordyceps spp.) 中一種獨特的藥用真菌。因其所含之多醣具有多種保健功效如抗發炎、抗氧化、抗腫瘤、抗病毒、免疫調節等功能,而受到市場重視。此多醣雖可透過液態培養取得,但其生產仍須經長時間發酵,因此本研究使用海藻酸鈣包覆蛹蟲草菌絲體進行固定化發酵,後藉此建立可重覆使用之生產系統,除了可提高蛹蟲草胞外多醣的產率,此可重覆利用之生產系統亦有利於工業生產。首先透過反應曲面法優化,可發現於優化條件下發酵至第 6 天時的胞外多醣產量為1.37 g/L,相較於未優化條件 (0.72 g/L) 可有效提高多醣產量達90.2%。且此系統能被重複使用至少三次,其中以第二次時的胞外多醣產量最高 (5.39 g/L)。後將此條件用於於氣泡管柱反應器生產,可發現於第二批次之第 15 天具有最大產量約10.42 g/L。所得之胞外多醣組成主要以以莫爾數比 8.91:0.09 之葡萄糖與半乳糖所組成。其抗菌性則是對 Staphylococcu saureus 具有最小抑菌濃度達 200 mg/mL。於抗氧化性上,對 DPPH 自由基和 ABTS 自由基之 IC50 分別為 4.65 mg/mL 和 7.92 mg/mL。於細胞毒性評估上,小於 150 mg/mL 濃度內不會對小鼠纖維母細胞 NIH-3T3 產生毒性。因此本研究能有助於蛹蟲草胞外多醣建立具經濟效益之工業生產模式,並有助於其應用於食品或醫療產業。
