Abstract
  停經婦女常伴隨肥胖,特別是腹部脂肪累積,也因此增加罹患代謝症候群之風險。許多飲食因子已被指出具抗肥胖之作用,其中十字花科蔬菜衍生物Indole-3 carbinol (I3C) 和洋蔥可降低因飲食誘導之肥胖,然而對於因停經造成的脂肪堆積影響尚不明確,因此本研究以卵巢切除 (Ovariectomized, OVX) 動物模擬停經狀態,探討給予I3C和洋蔥對OVX所誘發肥胖之影響。60隻雌性C57BL/6小鼠餵予含53% 脂肪的高脂飲食,隨機分為六組,包括假手術SHAM組、OVX組,以及OVX小鼠分別介入5% 洋蔥、0.1% I3C、混合5% 洋蔥及0.1% I3C和於飲水添加0.1% S-methyl-L-cysteine (SMC) 的組別,介入15週後以micro-CT檢測脂肪和肌肉組織體積,並於第16週犧牲。結果顯示,實驗期末OVX組體重雖與Sham組相似,但OVX顯著誘導停經後脂肪堆積和肌肉減少之現象,包括降低子宮重量、血清雌激素濃度與肌肉體積並增加腹部脂肪重量、脂肪體積及腹部脂肪脂質合成酵素Fatty acid synthase (FAS) mRNA表現。而在不影響攝食量情況下,攝取洋蔥的體重雖較OVX組增加,但能降低總脂肪重量並增加肌肉體積、雌激素合成酵素CYP19以及棕化指標PR domain containing 16 (PRDM16) mRNA與Uncoupling protein 1 (UCP 1) 蛋白質表現,且和其活性組成SMC皆能降低OVX後增加的腹部脂肪重量及FAS表現。綜合以上結果顯示,飲食中介入洋蔥可抑制OVX所造成之腹部脂肪堆積,此作用可能與其增加雌激素生合成酵素表現,促進白色脂肪細胞棕色化以及所含的SMC抑制脂質合成,降低白色脂肪堆積有關。
