Abstract
Growth factor receptor bound protein 7(GRB7)屬於Grb family的成員之一,是一種銜接蛋白(Adaptor protein)。GRB7本身具有五個功能性區域(Functional domain),會參與許多訊息傳遞路徑[1]。
Human epidermal growth factor receptor 2(HER2)第二型人類表皮生長因子接受體二,是乳癌的分類其中之一[2],而至目前的研究裡,HER2和GRB7具有高度相關[3]。為了進一步探討GRB7和HER2進一步關係,我們利用了RT-PCR、Real-time PCR,針對不同的乳癌細胞株及臨床病人檢體(共213組)加以分析,發現無論是在乳癌細胞株或是臨床病人檢體中,HER2 positive的組別中,GRB7也會高度表現。我們也利用SKBR3這株乳癌細胞株,發現GRB7在正常狀態下,GRB7不只存在細胞質也存在於核內,但GRB7並不是藉由入核直接調控HER2的基因表現。此外,我們也構築了Knockdown GRB7 plasmid及Overexpression GRB7 plasmid,發現GRB7被抑制後,HER2 的表現也隨之降低;過度表現GRB7後,HER2的蛋白表現並沒有受影響。
利用天然化合物-薑黃素(Curcumin)針對乳癌細胞株SKBR3 進行加藥試驗,我們發現在24小時內,Curcumin可以大幅降低GRB7的蛋白表現量。進一步使用Cycloheximide加以分析,結果顯示加入Curcumin對於GRB7的抑制,可能是使GRB7蛋白走向被水解的路徑,而不是蛋白被抑制在合成的時期。希望能加以釐清GRB7蛋白被降解的確切機制,期盼在未來能藉由影響GRB7進而抑制HER2的過度表現。
