Abstract
長片段基因定序(long-read sequencing)具有能夠讀取較長序列、提高連續序列片段的定序正確度、定序速度快、以及避免因PCR擴增出現的錯誤率及偏好性的優點。有別於僅能對片段基因序列進行定序的短片段定序平台,長片段基因定序近年來已開始廣泛運用在微生物的檢測,將微生物的分子診斷提升至全基因領域。近年來的研究發現了人體健康狀況與腸道微生物群的關聯性。在我們目前的試驗,使用納米孔長片段定序平臺和超高效液相色譜(Ultra Performance Liquid Chromatography,UPLC)耦合MS/MS串聯的質量分析器(Mass Spectrometer,MS)方法,鑑定糞便中的腸道微生物群和腸道代謝物,發現腸道微菌叢和腸道代謝物的組成與慢性腎病進程發展有關。根據以上這些研究發現了腸道環境影響腎功能損害的新興潛在途徑,藉由進行橫斷面研究,試驗結果發現在重度慢性腎臟病(chronic kidney disease, CKD)狀況下,腸道中K. pneumonia或S-Adenosylhomocysteine的豐富度(relative abundance)會明顯增加。由以上結果可以得知,腸道微生物群中K. pneumonia或糞便中的S-腺苷酰高半胱氨酸S-Adenosylhomocysteine微生物群或代謝物的組成或漸增。都將可做為預測潛在高風險慢性腎臟病(chronic kidney disease, CKD)病人的新穎性生物標誌。
