Abstract
在軟骨組織工程以及硬骨組織修復之相關研究中,目前仍有許多急待解決之課題。對軟骨組織工程而言,如何在體外快速並大量的增生具有組織活性的軟骨細胞;對硬骨組織修復而言,如何促進大部骨缺損之骨增生與植入物之骨整合,皆為重要且待解決之課題。本研究探討第二型膠原蛋白對軟骨細胞及中胚層幹細胞之影響,以期利用其特性於軟骨以及硬骨組織之修復與重建。
於本研究中,外加第二型膠原蛋白於大量連續分盤增生培養後之人類軟骨細胞,可刺激已去分化之軟骨細胞再度分泌軟骨性標誌蛋白。此效應之訊息傳遞路徑為第二型膠原蛋白與引起之FAK與ERK的磷酸化,並活化下游之SOX9蛋白,促成軟骨性標誌蛋白之再度表現。本研究亦探討第二型膠原蛋白對中胚層間葉幹細胞硬骨分化之影響。被覆第二型膠原蛋白之表面,不但對經過骨化培養基處理過之中胚層幹細胞有極佳的親和力,並且可加速貼附其上之中胚層幹細胞骨化分化的程度與鈣質沉積的能力。此效應為經由FAK-JNK之訊息傳導路徑與RUNX2的磷酸化。綜合而言,本研究探討第二型膠原蛋白於體外大量增生之軟骨細胞再活化之應用與分子機轉,亦證實其促進中胚層幹細胞骨分化與其調控機制。本結果也顯示了第二型膠原蛋白未來應用於軟骨組織工程以及作為硬骨組織修復材料之潛力。
