Abstract
鎘為土壤、岩石、空氣及水中常見的重金屬,對人體為非必需且有健康危害的元素。臺灣經濟逐漸發展使得環境受到衝擊,人類活動所產生之廢水會造成嚴重污染。鎘為桃園地區土壤的主要污染物,控制場址鎘濃度均超過臺灣土壤污染管制標準規定食用作物農地的管制標準值則為 5 mg/kg,曾爆發鎘米事件導致大範圍農地停止耕種,當地鎘的生物可及性高,對於人體毒性強有致癌的風險。當地居民可能會經由食入等方是暴露於鎘。生物可及性及生物有效性為藥物動力學的特性,近年來廣泛應用於整治土壤污染,透過體外模擬實驗和以動物作為人體模型進行評估土壤重金屬進入人體後實際會被吸收的比例,進而應用在健康風險評估模式中。
本次實驗採集8個場址之土壤樣本,進行土壤基本性質、鎘濃度、生物可及性體外試驗(簡化生物可及性萃取法SBET、體外生理基礎萃取實驗PBET)、序列萃取實驗,相對生物有效性實驗以管餵氯化鎘作為參考物質,並選出2個鎘濃度超過土壤污染管制標準之規定一般土壤鎘的管制標準值(20mg/kg)的土壤樣本進行動物實驗,以小鼠的肝臟及腎臟作為生物標誌。以線性迴歸探討生物可及性結果與土壤基本性質、鎘濃度、序列萃取各相態比例之間的相關性。
土壤的基本性質結果範圍:pH值4.38 - 5.64、有機碳含量12.84 - 17.28 %,含水率2.98 - 4.01 %,質地分別屬於壤土及砂質壤土,土壤鎘濃度範圍為6.23 - 36.82 mg/kg,生物可及性平均結果:SBET 82.24 ± 7.39 %、PBET胃相86.57 ± 8.20 %、PBET腸相35.90 ± 11.65%,序列萃取主要分布於第一相態及第四相態。相對生物有效性結果:肝臟19.58 ± 4.05 %、腎臟30.93± 4.96 %、肝臟+腎臟26.89 ± 5.54 %。以肝臟加腎臟作為生物標誌所得之值進行健康風險評估,與土壤鎘全量濃度計算得的危害商數相比,考慮生物可及性結果及相對生物有效性結果則危害商數均降低,所有風險計算數值均小於1,該鎘污染場址對於成人或孩童均沒有非致癌的健康風險。
進行人類健康風險評估時,納入生物可及性或相對生物有效性,能較精確估計鎘對於人體造成的健康風險。根據本次研究結果,需再蒐集更多土壤以確認不同體外試驗生物可及性結果與相對生物有效性結果之間的關係。
