Abstract
本研究主要目標在於探討無患子種子蒸餾液在傷口癒合過程之中具有之生物活性與醫療領域的應用。從無患子種子蒸餾液的製備開始,將其運用於抗菌、抗發炎以及組織修復等測試。以氣相層析質譜儀 (GC-MS) 進行植物化學成分分析發現無患子種子蒸餾液包含2種生物活性成分苯類:1,3-bis(1,1-dimethylethyl)與 2,4-di-tert-butylphenol (2,4-DTBP)。抗菌測試顯示粉水比1:2之無患子種子蒸餾液 (SMSD 1:2) 可減少60%的金黃色葡萄球菌菌落生成數(p < 0.05)。而體外試驗於無血清環境培養下,無患子種子蒸餾液可增加皮膚纖維母細胞CCD-966SK的細胞存活率,其中SMSD 1:2效果最顯著;對NRK-52E細胞也是如此。刻痕試驗中,無論在24 hr或48 hr處理時間之下,SMSD 1:2促進CCD-966SK遷徙並且促使其刻痕大幅癒合(p < 0.05)。抗發炎測試使用LPS刺激小鼠免疫細胞RAW264.7產生NO,而SMSD 1:2可以減少NO生成(p < 0.01);另一方面以LPS刺激NRK-52E細胞產生發炎反應,SMSD 1:2使細胞活性上升,保護細胞受到毒害,在同一濃度之LPS作用下,兩個組別的數值最大可相差32.90% (p<0.001)。動物試驗藉由觀察大鼠皮膚傷口癒合情況評估無患子種子蒸餾液的影響,SMSD 1:2幫助其表皮修復,相較於控制組,傷口於第14天明顯縮小範圍(8.25% ± 3.85% vs. 14.04% ± 3.50%, p < 0.05),組織切片也可觀察到相同的情形;另外以LPS刺激小鼠產生發炎反應,同時以無患子種子蒸餾液取代飲用水餵養,可觀察到小鼠腎臟組織的發炎狀況比僅施打LPS並飲用一般蒸餾水的小鼠腎臟組織輕微。經由上述這些發現,無患子種子蒸餾液具有傷口癒合的潛能,並且抗菌與抗發炎,可能可以作為皮膚保養的成分之一,也提供減緩腎臟慢性發炎一個潛在的天然應用材料。
