Abstract
藥物開發的工作動輒需要耗時十年以上,並且失敗率很高,往往投入大量經費與人力後面臨計畫終止。根據過去研究資料發現,臨床前試驗中的藥物動力學(Pharmacokinetics, PK)研究被指出是常見導致藥物開發失敗原因,約有40%的研究因藥物動力學問題而失敗。由於藥物動力學通常是以動物實驗為主,透過反覆實驗來確保臨床試驗的有效性和安全性,除了會消耗大量實驗時間與成本之外,與人類的藥物動力學相關性之不確定性也可能會導致後續的問題。近年來機器學習逐漸興起,為加快研究步調,科學家們開始投入利用機器學習來預測藥物動力學特性。早期研究大多使用單任務模型做預測,研究預測的藥物動力學特性也較少,近年研究逐漸轉向多任務學習模型來提升準確度,然而可公開取得的資料並不多,且藥物動力學特性的涵蓋度也不足。
基於上述觀察本研究欲建立一個涵蓋完整人類藥物動力學特性的多任務學習模型以協助藥物設計,並探討資料特性與多任務學習的相關性。本研究首先收集並整合兩個公開資料集,一共包括10種人類藥物動力學特性的資料,接著以隨機森林與基於隨機森林的多任務學習演算法MTForestNet進行單任務與多任務的模型研究。
研究結果表明,基於結構指紋的模型,與採用同特徵的單任務隨機森林模型相比,多任務模型均方根誤差的平均值提升超過8.5%。而在個別任務中,以平均停留時間的均方根誤差改善幅度最大,達到16.30%。其中也發現到改善幅度較多的任務資料分佈幾乎為常態分佈,顯示此分佈的資料較能互相學習進一步提升準確度。研究也發現即便資料中有非常態分佈任務,其他任務也能從中學習到相關性,提升自身預測結果。最後模型結合適用域分析,進一步提升準確度,並為未來實際應用上做好準備。本研究建立了一個用於人類PK特性預測的多任務學習模型,提供10個PK特性以供預測,配合ECFP 特徵與常態分佈任務有不錯的預測能力,期待未來整合更多樣本數之後,能成為藥物開發過程中的一大助力。
