Abstract
本研究發展出利用Ganoderma tsugae合併漂白劑NaClO2的一鍋製成法,可快速且有效率的製造並萃取sacchachitin (SC)並且有很好的成效。進一步將SC利用NanoLyzerR做機械性奈米化以及TEMPO的氧化自由基反應做化學性奈米化,生成的材料利用FT-IR, solid state NMR, X-ray diffraction, carboxylate 含量測定, size和zeta potential做化學性質分析,物理性質如成膠特性、吸水特性來加以評估。實驗結果指出利用NanoLyzerR流經五循環所產生的SCN5,以及利用3.33以及5.00 mmol-NaClO/g-SC的TEMPO氧化反應所產生的奈米化材料分別命名為T033SC和T050SC,有著很好的發展潛力。將SCN5, T033SC, 和T050SC分別以2% (w/v)凍乾所形成的膜具有高孔洞性。在傷口癒合的應用上,SCN5和T050SC結合2-Acrylamido-2-methyl-1-propanesulfonic sodium (AMPs)為主架構的水膠,可以促進慢性傷口癒合並利用組織學評估也有很好的成效。在骨再生的應用,利用microCT評估,T033SC可以有效的促進骨癒合。綜合以上的結果,證明這新發展出的生醫材料在慢性傷口癒合及骨再生具有極大的應用潛力。
